# Construct QC linter

Lint a coding DNA sequence for premature stops, internal RBS/polyA motifs, unwanted restriction sites, GC extremes and repeats.

Batchable via `POST /batch`.

## Request body

- object
  - `sequence` string, required: Nucleotide sequence (raw or FASTA; IUPAC accepted).
  - `frameStart` integer: 1-based nucleotide where the reading frame begins.
  - `avoidEnzymes` string[]: Enzyme names whose internal sites should be flagged as errors. Matched against the curated common-enzyme set plus the Golden Gate Type IIS enzymes (BsaI, BbsI, Esp3I/BsmBI); an unrecognized name is rejected, never skipped.
  - `gcWindow` integer: Sliding-window size (nt) for GC-extreme scanning.
  - `gcLow` number: GC% below this flags an AT-rich window.
  - `gcHigh` number: GC% above this flags a GC-rich window.
  - `homopolymerMin` integer: Minimum run length to flag a homopolymer.
  - `crypticOrfMinAa` integer: Minimum peptide length (aa) for a hidden alternate-frame ORF to be flagged.

## Other responses

- `400`: Invalid argument or the tool could not run.
- `404`: Unknown tool.
- `429`: Rate limited (metered tools only). Body carries code `rate_limited` and retryable `true`; honor the Retry-After header.
- `503`: A metered tool is momentarily at capacity. The identical request should succeed after Retry-After; body carries code `upstream_unavailable`.
